چکیده
Exit from multipotency and lineage commitment of mesenchymal stroma cells (MSC) depends on microenvironmental cues the stem-cell niche but steering cell fate into desired in vivo remains a challenge. Increasing evidence suggests that glycosaminoglycans (GAGs) can be used to activate or sequester growth factors with specific action depending sulfation levels. It is postulated differentially sulfated biomaterials aid developmental instruction MSC guide tissue morphogenesis vivo. By application new injectable TGFβ-loaded heparin-PEG-hydrogel authors here gain ability control skeletal down chondral versus endochondral pathway status. High allows for long-term TGFβ-retention silencing Hedgehog-, BMP-, WNT-pathways installing prochondrogenic antihypertrophic which permitted permanent, collagen-type-II-rich neocartilage resistance calcification bone formation. Selective 6-O/N-desulfation heparin supports Hedgehog/BMP/WNT-signaling switching differentiation strong hypertrophic/osteogenic marker expression calcification. This work identifies GAG as crucial signal determine via prohypertrophic pathways, providing first proof-of-principle how modification-patterns lineage-choice during

Stem cell proliferation versus meiotic fate decision in Caenorhabditis elegans.
The C. elegans germ line has emerged as an important model for -understanding how a stem cell population is maintained throughout the life of the animal while still producing the gametes necessary for propagation of the species. The stem cell population in the adult hermaphrodite is relatively large, with stem cells giving rise to daughters that appear intrinsically equivalent; however, some of...
متن کاملSimultaneous regulation of miR-451 and miR-191 led to erythroid fate decision of mouse embryonic stem cell
Objective(s): Various microRNAs (miRNAs) are expressed during development of mammalian cells, when they aid in modulating gene expression by mediating mRNA transcript cleavage and/or regulation of translation rate. miR-191 and miR-451 have been shown to be critical regulators of hematopoiesis and have important roles in the induction of erythroid fate decision. So, the aim of this study is inve...
متن کاملHybrid Protein–Glycosaminoglycan Hydrogels Promote Chondrogenic Stem Cell Differentiation
Gelatin-hyaluronic acid (Gel-HA) hybrid hydrogels have been proposed as matrices for tissue engineering because of their ability to mimic the architecture of the extracellular matrix. Our aim was to explore whether tyramine conjugates of Gel and HA, producing injectable hydrogels, are able to induce a particular phenotype of encapsulated human mesenchymal stem cells without the need for growth ...
متن کاملImproving the Growth Rate of Human Adipose-Derived Mesenchymal Stem Cells in Alginate/Gelatin Versus Alginate Hydrogels
Background: Expansion and differentiation of stem cells relies on the soluble materials as well as the physical conditions of their microenvironment. Several methods have been studied in attempt to enhance the growth and differentiation rates of different adult stem cells extracted from different sources. Objectives: The purpose was to improve the three-dimensional (3D) culture condition of th...
